Stopping disease at its gateway: the mouth

Praetorian Bio is pioneering a new class of preventive biologics that target the pathogens driving cavities and gum disease. By combining cutting-edge computational modeling with decades of translational research, we’ve identified precise antigen targets—such as Streptococcus mutans and Porphyromonas gingivalis—that link oral health to systemic disease.

First-in-class vaccine

Praetorian Bio is developing a first-in-class vaccine targeting the microbial drivers of dental cavities at their root. The approach aims to generate broad mucosal immune responses that provide durable protection against complex, biofilm-associated pathogens including Streptococcus mutans. A schematic overview of the mechanism is provided below.

BALB/c mice were intranasally immunized on Days 0 and 21 with different vaccine formulations. All included a potent mucosal adjuvant, except Concept B, which was also tested without adjuvant. Two weeks post-boost, serum and nasal washes were collected and reactivity against whole-cell Streptococcus mutans was quantified by ELISA.

Induction of systemic and mucosal immune responses against S. mutans

All vaccine candidates induced strong systemic IgG and IgA responses. The absence of adjuvant markedly reduced antibody titers, confirming the adjuvant effect. Most importantly, all candidates also elicited S. mutans–specific mucosal IgA in nasal washes.

Why Praetorian Bio is Different

Precision

We target Streptococcus mutans and Porphyromonas gingivalis with highly specific antigens — delivering protective immune responses without disrupting the beneficial oral microbiome.

Prevention

Our vaccines stop pathogens at their point of entry — the mouth — preventing cavities, gum disease, and their systemic consequences before they take hold.

Scale

From design to manufacturing, our pipeline is built with scalability, manufacturability, and regulatory alignment in mind, ensuring global accessibility.
